    Landscape Genetics of the California Tiger Salamander: Inferences from Multiple Methods

    Landscape genetics is a rapidly growing field of study that compares patterns of gene flow among populations with habitat heterogeneity across a landscape to infer the interaction between dispersal of individuals and their physical environment. Empirical data generated from a landscape genetics study can inform conservation and management strategies, making the field increasing popular. However, concerns have arisen in the literature that the field is expanding faster than the analytic framework that supports it. Multiple methods for generating estimates of the association among habitat types and dispersal (i.e., least-cost paths and resistance surfaces) have been proposed, and there is a debate as to which statistical methods are best for examining the genetic structure on a landscape. We use an integrated empirical- and expert-opinion-based strategy to generate a landscape resistance surface for the California tiger salamander, Ambystoma californiense, which is a species of conservation concern. We utilize several alternative analysis methods (e.g., CCA, MRDM, ResistanceGA, GESTE, and partial Mantel tests) to look for agreement among methods describing the relationship of landscape features and genetic variation. Our analysis revealed variation among methods for describing genetic structure in this A. californiense metapopulation, but all methods indicated the presence of genetic structure, to some extent, across the landscape. This empirical data set provides both a perspective on habitat management for A. californiense and on the suitability of several novel analysis strategies for landscape genetics

    Australian media's use of Facebook postings to report events of national interest

    Online social networking sites such as Facebook have grown exponentially in recent times, yet little research has examined how the mainstream news media use the information available on these sites. This study explores how the Australian media used the social networking site Facebook in reporting three different news events: the disappearance of Australian backpacker Britt Lapthorne; the death of 4-year-old Darcey Freeman; and the devastating \u27Black Saturday\u27 Victorian bushfires. Sixty-four articles from Australian newspapers were identified pertaining to these three case studies within a seven month period from August 2008 to February 2009. An inductive thematic approach was used to identify the way in which information from Facebook was utilised by journalists within these news stories. Three main methods of utilising information from Facebook were established: the reporting of group activity to gather information, discuss developments, and gauge general public sentiment; the use of profiles to report the lives of newsworthy individuals via their postings; and responses via Facebook groups and profiles of the specific reaction of families, friends and the general public to an event. The rise of social networking presents new challenges for journalists in relation to how they use information ethically and responsibly, and the privacy implications associated with media reporting of postings on social networking sites are discussed

    Large-Scale User Modeling with Recurrent Neural Networks for Music Discovery on Multiple Time Scales

    The amount of content on online music streaming platforms is immense, and most users only access a tiny fraction of this content. Recommender systems are the application of choice to open up the collection to these users. Collaborative filtering has the disadvantage that it relies on explicit ratings, which are often unavailable, and generally disregards the temporal nature of music consumption. On the other hand, item co-occurrence algorithms, such as the recently introduced word2vec-based recommenders, are typically left without an effective user representation. In this paper, we present a new approach to model users through recurrent neural networks by sequentially processing consumed items, represented by any type of embeddings and other context features. This way we obtain semantically rich user representations, which capture a user's musical taste over time. Our experimental analysis on large-scale user data shows that our model can be used to predict future songs a user will likely listen to, both in the short and long term.Comment: Author pre-print version, 20 pages, 6 figures, 4 table

    Combined Unilateral Blockade of Cholinergic, Peptidergic, And Serotonergic Receptors in The Ventral Respiratory Column Does Not Affect Breathing in Awake Or Sleeping Goats

    Previous work in intact awake and sleeping goats has found that unilateral blockade of excitatory inputs in the ventral respiratory column (VRC) elicits changes in the concentrations of multiple neurochemicals, including serotonin (5-HT), substance P, glycine, and GABA, while increasing or having no effect on breathing. These findings are consistent with the concept of interdependence between neuromodulators, whereby attenuation of one modulator elicits compensatory changes in other modulators to maintain breathing. Because there is a large degree of redundancy and multiplicity of excitatory inputs to the VRC, we herein tested the hypothesis that combined unilateral blockade of muscarinic acetylcholine (mACh), neurokinin-1 (NK1, the receptor for substance P), and 5-HT2A receptors would elicit changes in multiple neurochemicals, but would not change breathing. We unilaterally reverse-dialyzed a cocktail of antagonists targeting these receptors into the VRC of intact adult goats. Breathing was continuously monitored while effluent fluid from dialysis was collected for quantification of neurochemicals. We found that neither double blockade of mACh and NK1 receptors, nor triple blockade of mACh, NK1, and 5-HT2A receptors significantly affected breathing (P ≥ 0.05) in goats that were awake or in non-rapid eye movement (NREM) sleep. However, both double and triple blockade increased the effluent concentration of substance P (P \u3c 0.001) and decreased GABA concentrations. These findings support our hypothesis and, together with past data, suggest that both in wakefulness and NREM sleep, multiple neuromodulator systems collaborate to stabilize breathing when a deficit in one or multiple excitatory neuromodulators exists

    (Non)Parallel Developmental Mechanisms in Vertebrate Appendage Reduction and Loss

    Appendages have been reduced or lost hundreds of times during vertebrate evolution. This phenotypic convergence may be underlain by shared or different molecular mechanisms in distantly related vertebrate clades. To investigate, we reviewed the developmental and evolutionary literature of appendage reduction and loss in more than a dozen vertebrate genera from fish to mammals. We found that appendage reduction and loss was nearly always driven by modified gene expression as opposed to changes in coding sequences. Moreover, expression of the same genes was repeatedly modified across vertebrate taxa. However, the specific mechanisms by which expression was modified were rarely shared. The multiple routes to appendage reduction and loss suggest that adaptive loss of function phenotypes might arise routinely through changes in expression of key developmental genes
